Purpose: To observe the effect of hyperthermia and radiation combined hyperthermia on apoptosis and Bcl-2 expression in CNE-2 cells line.thereby elucidating the antitumor mechanism of this combined treament.Methods:The cells were invided into four groups:normal group , radiation group,hyperthermia group,radiation combined hyperthermia group. The apoptosis rate and Bcl-2 expression rate were assayed by TUNEL and immunocytochemistry techniqnean.Result: All the groups could induce the apoptosis in CNE-2 cells line. With the increase in the doses of radiation,the apoptosis rate was elevated. It's easy to see the the apoptosis rate in radiation combined hyperthermia group was significantly higher than that of radiation group and hyperthermia group(P<0.05).The last three groups all declined the Bcl-2 expression in response to the doses of radiation(P<0.05). The expression of Bcl-2 was significantly lower than that of radiation group and hyperthermia group(P<0.05). The expression of Bcl-2 was contrary to the apoptosis rate in radiation group(P<0.05).But there was no statistical correlation in radiation combined hyperthermia group(P>0.05).Clonclusion: Radiation,hyperthermia,radiation combined hyperthermia may all induce apoptosis and cause reduction of Bcl-2 expression in CNE-2 cells line. Hyperthermia has been shown obvious radiosensitizing effect.
