Ischemic cerebrovascular disease is one of common clinical diseases with high incidence, mortality and disability. It has posed a serious threat to human health and has become one of the three major causes of death. The morbidity in our country is high. Traditional Chinese medicine has traditional advantage on the treatment of cerebral infarction, Enhancing-qi and activating-blood and clearing- phlegm Method is one of basic treatments on cerebral infarction Part One :Documents and theoretical studyReviewing the path-physiological mechanisms and other relevant factors of the ischemic cerebrovascular disease.The path-physiological mechanism of cecebral infarction is complicated, which includes physiological self-regulation, formation of penumbra and reperfusion damage. Severe decompensation after cecebral infarction involves in calcium over-loading, excitatory damage, radical free damage, inflammation reaction, apoptosis, and so on. Modern medicine treatment consists of basic treatment, dissolving thrombus treatment, anticoagulation, neuroprotection and rehabilitation.Reviewing pathology and pathogenesis of stroke, the dissertation emphasized on wind, fire, deficiency, retention of phlegm and fluid and blood stasis on the pathogenic mechanism of stroke. The dissertation also summarized the study progress on ischaemic stroke of Chinese medicine treatment in recent years.The relationship of qi , blood and body fluid, the stagnation of phlegm and blood due to qi deficiencyis is the main pathology and pathogenesis of ischaemic stroke. Enhancing-qi and activating-blood and clearing- phlegmMethod treat the recovery period of ischaemic stroke. Perhaps the essence of stagnation of phlegm and blood of ischaemic stroke is hypercholesterolaemia. The explanation of Fufanghaidai Capsule. Part Two:Experimental research ObjectiveThrough the selection experiment to establish three materials' extraction techniques and Consumption ratio.To study the effects of Fufanghaidai Capsule on hemorheology^ micro blood circulation and experimental thrombosis form of the blood stasis rats model. The rat model of cerebral ischemia was established by middle cerebral artery occlusion (MCAO) with nylon suture. To study the protective effects and the mechanism of Fufanghaidai Capsule on experimental cerebral ischemia in rats. Methods : (l)Three selection experiments:To select by two experiments: (I) the effects of shaixuanfang on the neurological deficit score of the Cerebral Ischemia mice model;(2)the effects of shaixuanfang on the survival time and gasping times of isolated heads of the Cerebral Ischemia mice model;(2)Experimental study on Pharmacodynamics of Fufanghaidai Capsule to the blood stasis rats model.To study the effects of Fufanghaidai Capsule on hemorheology> micro blood circulation and experimental thrombosis form of the blood stasis rats model.60 SD rats were randomly divided into six groups:normal group, model group, Fufanghaidai Capsule low, middle and high dosage groups, Fufangdanshendiwan group. The rats model of the deficiency of qi and blood stasis was made by injecting large dose epinephrine into rat and then immersing the rat in ice —water to make the rats in deficiency of qi and blood stasis situations. The blood viscosity^ plasma viscosity^ length> green weight and dry weight of thrombosis in vitro of different group rats were tested by the MS/XW-E hemorheology ^ micro blood circulation parameters checking device, Laser Doppler hemorheology monitors and other equipments.(3)The protective effects of Fufanghaidai Capsule on the rat model of cerebral ischemia was established by middle cerebral artery occlusion (MCAO) with nylon suture.The mail SD rats were randomly divided into seven groups: false operation group, model group, Fufanghaidai Capsule low, middle and high dosage groups, Fufangdanshendiwan group and Nimodiping group.To study the effects of Fufanghaidai Capsule on the neurological scale, cerebral water content , cerebral infarcted volume , and pathological exmnination of cerebral tissues of the (MCAO) rats model. (4)The mechanism research of Fufanghaidai Capsule on the rat model of cerebral ischemia was established by middle cerebral artery occlusion (MCAO) with nylon suture.The mail SD rats were randomly divided into seven groups: false operation group, model group, Fufanghaidai Capsule low, middle and high dosage groups, Fufangdanshendiwan group and Nimodiping group. Results: (l)Three selection experiments:Haidai is extracted as original technique, Huangqi and Chuanxiong are extracted from water after mixed . The technique that the three raw materials are extracted by 20%~60% alcohol after mixed is the most desirable, identified parts II, III merger separation processes.The composition of Haidai5peiwufang Haidai : Huangqi: Chuanxiong =48 grams : 12 grams : 6 grams (2)Experimental study on Pharmacodynamics of Fufanghaidai Capsule to the blood stasis rats model:Fufanghaidai Capsule low^ middle, high group and Fufangdanshendiwan group could decrease blood viscosity of high, medial, low shearing rate as well as serum and plasma viscosity(p<0. 01, respectively),Fufanghaidai Capsule middle group' s effect is the best.The date suggest that length, green weight and dry weight of thrombosis in vitro of Fufanghaidai Capsule, low, middle, high group and Fufangdanshen diwan group were lower than the model group (p<0.01, respectively), Fufang haidai Capsule middle group' s effect is the best.(3)The protective effects of Fufanghaidai Capsule on the rat model of cerebral ischemia was established by middle cerebral artery occlusion (MCAO) with nylon suture.Fufanghaidai Capsule low, middle and high dosage groups, Fufangdanshendiwan group and Nimodiping group markedly decreased neurological deficit score (P< 0. 01, respectively) and cerebral infarcted volume(P< 0. 01, respectively), antagonized the increase of water in the brain tissue of the (MCAO) rats model (P< 0. 01, respectively). Nimodiping group' s effect is the best, Fufanghaidai Capsule middle group' s effect follow it. Fufanghaidai Capsule low, middle and high dosage groups, Fufangdanshendiwan group and Nimodiping group significantly improved pathological exmnination of cerebral tissues of the (MCAO) rats model. Nimodiping group' s effect is the best, Fufanghaidai Capsule middle group' s effect follow it.(4)The mechanism research of Fufanghaidai Capsule on the rat model of cerebral ischemia was established by middle cerebral artery occlusion (MCAO) with nylon suture.Fufanghaidai Capsule low, middle and high dosage groups, Fufangdanshendiwan group and Nimodiping group significantly reduced activities of LDH and content of MDA in serum(P<0. 01, respectively) . enhanced activity of SOD in serum(P< 0. 01, respectively), . Nimodiping group' s effect is the best, Fufanghaidai Capsule middle group' s effect follow it. Cone I us i ons:The results proved that Fufanghaidai Capsule can significantly improve hemorheological parameters and micro blood circulation of the deficiency of qi and blood stasis rats model, and can inhibit the form of thrombosis in vitro of the deficiency of qi and blood stasis rats model.Fufanghaidai Capsule have significant protective effects on the rat model of cerebral ischemia was established by middle cerebral artery occlusion (MCAO) with nylon suture.The protective mechanism may be related to decreasing the content of MDA and increasing the activity of SOD and decreasing cerebral water content.
