Objective: Tripterygium wilfordii Hook f(TWHF),a member of the Celastraceae family,is poisonous.Tripterygium Wilfordii Hook.f shows extensive clinical effect.The most frequent adverse reaction is damage of digestive system which can use for a long time.The largest adverse reaction is damage of genesis endocrine system used for a long time,a great deal of research about regarding TWHF as a antifertility drug has been done in recent years.However,many experts are suspect it's effect in genetics when it is used as antifertility and remedy.Using TWHF to male mice under antifertility-dosage groups,medium-dosage groups and remedy-dosage groups in this study, in order to detect TWHF 's genital system toxicity at different point of view which can provid rationale and evidence to use TWHF safely during the period of clinical treatment.Method:This essay introducts micronucleus of bone marrow,sperm deformity,chromosome aberration,analyze of synaptonemal complex and single cell gel electrophoresis, approaching effect of TWHF to male mice in genesis toxicity from whole to detail gradually at different point of view.Result:1.Negative groups and medium-dosage groups shows no statistical significance compared with antifertility-dosage groups,the other groups shows statistical significance one another in micronuclei assay of bone marrow;2.Experimental groups shows statistical significance compared with negative groups and positive groups in sperm deformity assay;3.Medium-dosage groups and positive groups shows statistical significance compared with other groups in variation of body weight;antifertility-dosage groups,positive groups and blank group shows no statistical significance compared with one another,the others shows statistical significance in variation of testicles weight/body weight;4.Medium-dosage groups shows no statistical significance compared with positive groups,the other groups shows statistical significance one another in analyze of synaptonemal complex;5.Positive groups shows no statistical significance compared with medium-dosage and remedy-dosage groups,the other groups shows statistical significance one another in chromosome aberration;6.CASP image analyze of comet assay shows,experimental groups shows statistical significance compared with negative groups and positive groups,inter-target shows different slightly.Conclusion:1.Every-dosage group of GTW cause sperm deformity. Micronuclei of bone marrow cell indicates 20㎎/㎏.d group and 30㎎/㎏.d group of GTW generate somatocyte inherent toxicity.2.
