The Study Of Genotoxic Damage And HSPs Level In Peripheral Blood Lymphocytes Of Workers Exposed To Coke Oven Emission

Objective: To explore the damage degree of DNA in lymphocytes of workers exposed to coke oven emission.Methods: 104 coke oven workers(oven top, oven side, oven bottom)in a steel factory were selected as occupational exposure group while 28 workers unexposed occupational carcinogenic substances as the control group,who were employed in control disease centre of a steel factory. Personal general information including age, gender, history of person, occupational exposure history and smoking status was elicited during questionnaire-based interviews. Peripheral vein blood was drawn from every subject. The HSP70 level was measured by Western blot, and the degree of DNA damage in the peripheral blood lymphocytes was evaluated separately by micronucleus assay and single cell gel electrophoresis assay. The result suggests HSP70 could protect cells from damage,the expression of HSP70 could protect cells from damage.Results:①There were no significant differences among those groups regarding age and tobacco(P>0.05).②Micronucleus rates in three groups were separately 2.72‰,1.88‰and 1.33‰. Micronucleus rates of high dose and low dose groups were significantly higher than those of the control group(P<0.01), high dose group﹥low dose group﹥control group.③There was significant difference in tail length,Olive Tail Moment of SCGE(lgχ±s)of occupational exposure workers compared with the control group(P<0.05).④Levels of HSP70 in peripheral blood lymphocytes in three groups were separately 1.19±0.76, 0.98±0.36 and 0.91±0.51. They increased significantly compared with that of the control, Conclusion: PAHs of the coke oven emission could result in increased level of micronucleus rates in peripheral blood lymphocytes, DNA and genetic damage.The HSP70 in lymphocytes and SCGE were influenced.
