Objective:To explore the biomarkers of lung cancer screening in coke oven workers by studying the relationship between PAHs exposure and DNA damage and DNMT1 and HDAC1protein.Methods:Through the investigation of a coking plant in the operation field,according to the production equipment and process,the enterprise of 178 male coking workers as the exposure group,including furnace,furnace side worker,furnace top worker and auxiliary workers;at the same time the control group selected 146 male water power plant.The general situation and occupational exposure history of the investigated subjects were collected through questionnaires.According to the occupational hygiene standard,the monitoring and sampling of the coking site were collected,and the morning urine and fasting venous blood of the subjects were collected.To measure the occupation environment of BaP and 1-OHPy in urine by high performance liquid chromatography;according to DNMT1,HDAC1 ELISA kit for determination of DNMT1 and HDAC1protein content;DNA damage by comet assay.The Olive tail distance is analyzed with Comet IMI software after collecting images with a fluorescence microscope.Results:The content of benzopyrene in the workplace of contact group was higher than that of the control group,and the difference was statistically significant(P<0.05).The level of urine 1-OH-Py,the damage of DNA and the expression of DNMT1 and HDAC1 in the blood of the contact group were all higher than those in the control group,the difference was statistically significant(P<0.01).The expression level of DNMT1 and HDAC1 increased with the increase of internal exposure level,and the difference was statistically significant(P<0.01).The DNMT1,HDAC1 and OTM values of the smokers in the contact group and the control group were significantly different from those in the non smokers(P<0.01).The DNMT1 and HDAC1 protein expression levels of the drinking groups in the contact group and the control group were not significantly different from those in the non drinking group(P>0.05).The OTM value of the drinking group was compared with the non drinking group,the difference was statistically significant(P<0.05).In a word,DNMT1,HDAC1,1-OH-Py,smoking and drinking have an impact on the damage of DNA,and the optimal regression equation of DNA damage is Y=20.551+0.268X1+0.189X2+0.623X3+0.387X4+0.091X5,which determines the coefficient R2=0.68.Conclusions:Occupational exposure to coke ovens will cause DNA damage;DNMT1,HDAC1,1-OH-Py,smoking and drinking will all affect the damage of DNA;the serum DNMT1 and HDAC1 proteins can be used as a marker for early screening of lung cancer in coke oven workers.
