Objective Raising atherrosclerosis model, to observe the the local expression of angiotensin II (Ang II ), transforming growth factor-β1 (TGF-β1),plasminogen activator inhibitor-1 (PAI-1) and type IV collagen in renal tissue and to approach the renal protective effect of Captopril and Valsartan through blocking renin-angiotonin system(RAS) on various level. Methods Thirty male New Zealand white rabbits were divided into four groups randomly:(A group)cholesterol group, (B group) cholesterol+ Captopril treated group,(C group) cholesterol+Valsartan treated group and (D group)control group. After 10 weeks, take suction rabbit central artery empty stomach blood preparation and determin the level of total cholesterol(TC),triglyeride(TG) and low-density lymphocyte(LDL) using BackmaanLX20 automatic biochemistry analyzer. AngII level of plasma was evaluated by radioimmunoassay. The expression of Ang II, TGF-β1, PAI-1 and type IV collagen in renal tissue were examined by immunohistochemisrey.Results The expression of AngII,TGF-β1, PAI-1 and type IV collagen in renal tissue all concentrated on glomerular parietal epithelial cell,Mesangial cell,renal tubular epithelial cell and tubules interstitial substance area. The expression of AngII of renal tissue in A group increased significantiy compared to D group (0.391±0.049 vs 0.216±0.025,P<0.0001),captopril reduced AngII significantiy (0.232±0.027 vs 0.391±0.049, P<0.0001 ) and there is no significant deviation compared to D group (0.232±0.027 vs 0.216±0.025, P=0.28 ), valsartan did not have effect on AngII (0.379±0.035 vs 0.391±0.049, P>0.05) . Compared with Dgroup,TGF-β1 level of renal tissue in A group increased significantiy (0.439±0.045 vs 0.231±0.019, P<0.0001 ), Captopri and Valsartan decreased the expression of TGF-β1 respectively (0.245±0.062 vs 0.439±0.045,0.250±0.044 vs0.439±0.045, P<0.0001 ) .And there were no difference between B group and D group, C group and D group, and B group and C group (0.245±0.062 vs 0.231±0.019, 0.250±0.044 vs 0.231±0.019, P>0.05) (0.245±0.062 vs 0.250±0.044, P=0.691) . Compared with D group, PAI-1 level of renal tissue in A group increased significantiy (0.389±0.065 vs 0.196±0.017, P<0.0001), Captopri decreased PAI-1 (0.226±0.018 vs 0.389±0.065, P<0.0001 ) , but the level of PAI-1 was still taller than that of D group (0.226±0.018 vs 0.196±0.017, P=0.035), Also Valsartan decreased PAI-1 (0.222±0.017 vs 0.389±0.065, P<0.0001 ) ,and there were no significant difference between the level of PAI-I in C group and D group (0.222±0.017 vs 0.196±0.017, P=0.067). Compared with D group, type IV collagen level of renal tissue in A group increased significantiy( 0.409±0.037 vs 0.239±0.040,P<0.0001 ), there were no difference between B...
