Make the arrhythmia animal model by injected different concentration BaCI2 0.3%, 0.5%,0.8%,1.0%. 2:choose the healthy Guinea Pigs and assign 6 groups in blind, Promethazine A (0.312mg/kg) B (0.462mg/kg) C (0.693mg/kg) D (0.900mg/kg) E (1.400mg/kg) F (1.850mg/kg), which be equal to adult does A (25mg/60kg) B (50mg/60kg) C (75mg/60kg) D (100mg/60kg) E (150mg/60kg) F (200mg/60kg). After giving different does drugs, observe the PP,PR,QT interval change in eight time point 0,1,3,5,10,20,30,40 minute. 3:observe the antiarrhythmia effects of the Promethazine on arrhythmia Guinea pigs, with the NS as black control and Lidocaine as positive control. The index is the duration of arrhythmia.Result:The anaesthazine Guinea pigs of 4 groups both induced ventricular arrhythmia by injected different concentration BaCI2 with same speed. No arrhythmia occur in NS group, there is no statistic different in the arrhythmia occur time, but the duration has the statistic different in every two groups.In the study of Promethazine influence on the normal ECG of guinea pig at A,B,C,D groups, increased the concentration of Promathazine, the PP, PR, QT interval has the increase tendency. There is no statistic different between in 0 minute and 3,5,10,20,30,40 minute. This testified that the Promathazine hasn't influence on normal ECG in the treatment range. In the group E, Compared 1,3,5,10 minute with 0 minute, the PP, QT interval has the statistic different. In the group F, after giving drug, the Guinea Pigs suffer arrhythmia in 1 minute, Compared with 0 minute, The PP,QT interval has the statistic different in 3,5,10,20,30,40.This testified that Promathazine over the treatment range can slower the heart rat and induce arrhythmia. In the E, F group, the PP, QT interval different between 0 and 3,5,10,20,30,40 minute has statistic different with the A, B, C, D group, and the Promathazine of E, F group has the dramatic influence on the PP,QT interval than other groups.In the antiarrhythmia study.
