Objective To study changes of DNA content in bosom bone cells of human with the postmortem interval, trying to provide objective support for estimating of PMI.Methods 15 bosom bone of human corpses,whose PMI and constitution and causes of death were known were collected. Using Methyl green Pyronine B staining methods and HE staining methods of DNA and the computerized image analysis to measure the DNA content in bosom bone cells of human during 7d after death.Results There was a certainty relationship between the degradation rate of the bosom bone DNA and the PMI(in 7d) of human.Three parameters reflecting the degradation,including integral optical density, average gray and aimed area ratio were related to PMI significantly.Conclusion By the way of Methyl green Pyronine B staining methods and HE staining methods of DNA and the computerized image analysis,we can draw two conclusions. 1 ,the DNA degradation in bosom bone cells of human corpses can be used as a target to estimate the PMI.2,we can measure the DNA content in bosom bone cells of human corpses to estimate the PMI during 7d after death.
