Objective: To investigate the relationship between the renal microvascular injury and progressive renal failure in remnant renal tissues of 5/6 Nephrectomy young rats,and evaluate the correlation between them, and study the interfering effects of treated with angiotensin converting enzyme inhibitor(ACEI) and angiotensin receptor angonist(ARB) .Methods: The remnant kidney is a representative model in the study of progressive renal disease. When the models were succeed, then, those animals were divided randomly into three groups: sham group; untreated group and treated group by ACEI combined with ARB.. At 2th, 4th ,8th , and 12th week, we measured the body weight, blood pressure and proteinuria/24h serum biochemical parameters, renal pathological changes and detected the expression of VEGF/TSP-1 protein which reflecting the microvascular in renal tissues by the way of immunohistochemical technique. These protein expressing changes were evaluated and to analysis the pathogenitic role of these parameters changes in renal lesions, and to evaluate the interfering effects of treatment with ACEI andARB.Results: Urinary protein, serum biochemical parameters were increased progressivelythe, body weight were not increase or increase slowly in untreated group. However, in treated group, these changes were ameliorated following treated with ACEI and ARB.At all time points in experiment, each parameters, such as serum biochemical, proteinuria et al are normality in sham group rats. The renal pathological results by HE staining at 12th week had showed that mesangial proliferated and its matrix to broad, the mass fibrin accumulated in Borman's capsule, glomerular capillaries were collapsed, tubule were dilatalted or atrophied widespreadly, monocyts/mocrophages infiltrated into tubulointerstitial in untreated group. But those pathological changes was ameliorated significantly in treated groups. Through immunohistochemical staining, and semiquantitative analysis of immunohistochemical positive signal, we had detected that the protein expression of VEGF/TSP-1 in renal tissue .VEGF were decreased generally in untreated group, otherwise, TSP-1 were increased.In treated groups, both of them were ameliorated significantly at corresponding time points.
