Objective: Mainly to observe the neuropeptide Y Yl receptor immunoreactive neuro, detect the changs of the expression of NPY Yl receptor's (NPY YlR) protein and the concentration level of serum follicle stimulating hormone (FSH) and.luteinizing hormone (LH) after leptin injected into lateral ventricle of ovariectomized but estrogen primed (OEP) mice, in order to approach the mechanism of leptin regulation the reproduction function, and provide some foundation experiment date for leptin's clinical application.Methods: 60 adult female C57BL/6J mice were used for OEP animal model: mice were bilateral ovariectomized in sterile condition, then 2μg of estradiol benzoate (E2) were subcutaneous injection (S.C.) per mice per day after 10 days' resting, which lasted 10 days, and we call it OEP mice. After that, all of the OEP mice were divided into treated group and control group, 4μg leptin dissolved in 2μl saline were microinjected into the lateral ventricle of the treated group mice, and 2μl saline without leptin were injected into the control group, both two groups were divided into 4 terms depend on the time 1.0h, 1.5h, 2.0h, 2.5h after lateral...
