Cell apoptosis is also named cell programmed death, which is one of thenewest researches in biology. Apoptosis was closely associated with manypathological and physiological conditions. It plays an important role in humanhealth.As far as 100 years ago, Carl Vogt found this process. Kerr et al. havenamed it as "apoptosis" at 1972. Apoptosis is an initiative cytology processregulated by different genes, such as Ced gene family, bcl -2 gene family,Fas/Apo gene family, P53 gene and ICE gene family etc.Evodiamine was isolated from the fruits of Evodia rutaecarpa var.bodinaieri huang. As one of Chinese traditional medicine, it has been used forcure disease for thousands years in China. Recently, evodiamine showed tumorsuppression activity. Evodiamine can induce apoptosis of human melanomacancer cell A375-S2, HeLa, MCF-7, THP-1, CCRF-CEM and L929.Our research is to elucidate the molecular mechanism of evodiamineantitumor activity. Caspase-3 and -8 were activated in the process ofevodiamine induced HeLa cell. But it didn't affect the activity of caspase-1. Weused flow cytometry, RT-PCR and western blot to determine the mechanism.We found cell was blocked at G2. The number of cell apoptosis that wasinduced was dependent with the concentration of evodiamine and reaction time.The p53 transcription and expression was increased, while bcl-2 was reduced.The expression of cyclin B1 was cut down and Bax was raised. It suggests thatevodiamine antitumor activity was related to p53, bcl-2, Bax and caspase.
