Studies On Chemical Constituents Of Saponins And The Method Of Determination Of Caulophyllum Robustum M.

Caulophyllum robustum Maxim which belongs to the family of Berberidaceae distribute in eastern north,Shanxi province,Gansu province,Sichuan province and far east of Russia mostly. Its officinal part was radix and rhizoma and it can be planted. The herb which was recorded in 《Chinese traditional medicine dictionary》 is a folk medicine that has already been utilized widely to cure many diseases such as rheumatoid arthritis, stomachache and bellyache,fracture injuries and menstruate disorder etc. due to its strong and wide biological activities. It contain alkaloids and saponins mainly according to literature.The experiment results confirmed that the part of extracted with 70% ethanol from Caulophyllum robustum.M had the effects of anti-rheumatism. Chemical and pharmaceutical research have been conducted on the part extracted with 70% ethanol from Caulophyllum robustum.M for the aims of further determining the active compounds and effect mechanism for anti-rheumatism. 7 compounds have been isolated with silica. gel, reversed-phase (ODS) column chromatography and HPLC from this part and the structures of 7 compounds have been fully determined according to the chemical and physicochemical evidence (1D and 2D-NMR). Among them,4 new compounds were firstly reported and 3 compounds were firstly found in Caulophyllum robustum.M. The structures of the four new compounds were identified as leiyemudanoside A[3-O-α-L-arabinopyranosyl- hederagenin-28- O-P-D-glucopyranosyl-(1→6)-β-D-glucopyranoside], leiyemudanosideB[3-O-β-D-glucopy-ranosyl-(1→3)-α-L-arabinopyranosyl- caulophyllogemn-28-O-α-L-rhamnopyrano-syl-(1 →4)-β-D-glucopyranosyl-(1 →6)-β-D-glucopyranoside],leiyemudanoside C[3 -O-β-D-glucopyranosyl-(1→2)-α-L-arabinopyranosyl-caulophyllogenin-28-O-α-L-rhamnopyranosyl-(1 →4)-β-D -glucopyranosyl-(1 →6)-β-D-glucopyranoside], leiye mudanoside E[3-O- β-D-glucopyrano syl-(1→3)-α-L-arabinopyranosyl-echinocys-tic acid -28-O-α-L-rhamnopyranosyl-(1 →4)-β-D-glucopyranosyl-(1 →6)-β-D-gluc-opyranoside] 。In order to control the medicinal materials's quality,this thesis still determined four constituents simultaneously by HPLC-ELSD for the first time. It has already been proved that the method which was simple,accurate, effecttive and feasible etc can be used for controlling quality of Caulophyllum robustum.M by optimizingchromatographic conditions of HPLC and Parameters of ELSD and seeing about precision,stability and repeatability.
