Caulophyllum is a small genus of perennialherbs in the family Berberidaceae,the root and rhizome of Caulophyllum robustum Maxim is the source of traditional Chin ese medicine “hong mao qi”.Mainly distributed in the northeast,Hubei,Tibet and oth er places,Its root and rhizome have been used to induce childbirth,ease the pain of labor,rectify delayed or irregular menstruation,alleviate heavy bleeding and pain durin g menstruation,and alkaloids,triterpene saponins,iridoids,fatty acids,sterols and othe r chemical components have been isolated from it.And found that anti-rheumatoid,an ti-inflammatory analgesic,antibacterial,anti-oxidation and anti-tumor activity.The compounds of C.robustum Maxim were isolated by column chromatography with silica gel,Sephadex LH-20 and ODS-HPLC.The structures were elucidated by o ptical?IR?MS?1D-NMR?2D-NMR analysis of spectroscopic data.fourty compounds were isolated and their structures were identified as 23-hydroxy-3,19-dioxo-olean-12-en-28-oic-acid(1),23-hydroxy-3,11-dioxo-olean-12-en-28-oic acid(2),16?,23-dihydroxy-3-oxo-olean-12-en-28-oic acid(3),collinsonin(4),Eechinocystic acid 3-O-?-L-arabinopyr anoside(5),Hederagenin(6),saponin PE(7),3-O-?-L-arabinopyranosyl-gypsogenin-28-O-?-L-rhamnopyranosyl-(1-4)-?-D-glucopyranosyl-(1-6)-?-D-glucopyranosyl estr(8),3-O-?-D-glucopyranosyl-gypsogenin-28-O-?-L-arabinopyranosyl-(1-6)-?-D-glucopyranosyl ester(9),3-O-?-L-arabinopyranosyl-gypsogenin-28-O-?-L-rhamnopyranosyl-(1-4)-?-D-glucopyr anosyl-(1-6)-?-D-glucopyranosyl ester(10),4,4'-Diphenylmethane-bis(methyl)Carbamate(11),Magnolamide(12),magnoflorine(13),maguocurarine(14),Columbamine(15),N-methylcytisine(16),baptifoline(17),2,4'-Diphenylmethane-bis(methyl)Carbamate(18),Obtucarbamate A(19),(-)-8,8?-dimethoxy-l-O-(?-D-glucopyranosyl)secoisolariciresin ol(20),Ligalbumoside A(21),Alangilignoside D(22),(-)-5?-methoxyisolariciresinol 3?-O-?-D-glucopyranoside(23),1-Linolenoylglycerol(24),1-Linolenoylglycerol(25),echi puroside A(26),?-hydroxypropiovanillone 3-O-?-D-glucopyranoside(27),(6S,9R)-rose oside(28),siringin(29),Cimifugin(30),Vanillin(31),p-hydroxybenzoic(32),p-hydro xyphenethylalcohol(33),Ferulic Acid(34),Isferulic Acid(35),?-hydroxypropiovanillone(36),p-coumalic acid(37),2-[4-(3-hydroxy-1-propenyl)-2-me-thoxyphenoxy]-1,3-propan ediol(38),benzoic Acid(39),rel-5-(1R,5S-Dimethyl-3R,4R,8S-trihydroxy-7-oxa-6-ox obicyclo-[3,2,1]oct-8-yl)-3-methyl-2Z,4E-pentadienoic acid(40).Compounds 1-3 were new Compounds;Compounds 4,6,8,11,12,14,15,18-40 were isolated from this genus for the first time.Lipopolysaccharide(LPS)induced RAW264.7 cells,Elisa reagent method is used to detect compounds NO inhibitory effect on cell secretion,evaluate compounds 1-10 of anti-inflammatory activity in vitro,the results showed that compounds 1-5 has inhib itory effect on the release of NO,and antitumor activity of A549 cells in vitro screen ing,the results showed the inhibition of compounds 1-5 on the A549 cells,the resear ch provides the foundation for the further development of C.robustum Maxim.
