Maize(Zea mays L.) is one of the traditional crops, which is widely cultivated in China. Stigma maydis, the by-product of maize, is greatly abundant, while it hasn't been made good use. A great deal of Stigma maydis has been wasted in vain except small parts' being used in medicine industry. Stigma maydis is of both obviously pharmacological value and certain character of dietetic treatment, hence it possesses broadly the prospect of study and development. Nowadays researchers have done much on the clinical application of Stigma maydis, but they did less on the extraction process of flavonoids, one of the active components in Stigma maydis, and its pharmacological function. To make the best of the Stigma maydis resource and exploit agricultural by-produce, in this research I took Stigma maydis as material and researched extraction process, separation and purification, the bioactivity of Stigma maydis flavonoids. The research results were as follows:1. The content of sugar in Stigma maydis was 1.12%, crude fat 3.57%, ash 6.54%. 15 mineral elements had been determined, including 9 essential elements among which the contents of Ca, Fe and Mg were higher. The contents of harmful elements Hg, Pb, As and Cd were lower. The content of 17 determined total amino acids added up to 11.02%, which included 7 essential amino acids accounting for 38.40% of total amino acids.2. Though the choice of the solvents for total flavonoids of Stigma maydis, ethanol was fitter to extraction. With the orthogonal experimental design, the optimum conditions of traditional extraction method was obtained at the solution of ethanol of 60%, extraction temperature of 60℃, solid-liquid rate of 1:20 and 3 extraction times per 2.0h.Under the conditions, the extraction ratio of total flavonoids from Stigma maydis were 1.786%.3. The optimum conditions of ultrasonical-assisted extraction were obtained as follows: ultrasonic power of 400W, ultrasonic time of 20min, the ethanol concentration of 60%, solid-liquid rate of 1:20, extraction temperature of 60℃and extraction time of 2.0h for one time.
