Franchet Groundcherry (Physalis alkekengi L.var. franchetii(Mast.) Makino) also name of Hong Gu-niang, is a kind of unique fruits, belong to Solanaceae family. The extraction process of polysaccharide in Phyalis calyx is optimized,thte crude polysaccharide was sepereted and purified, the structure of the purified polysaccharide was determined and simultaneous determination of crude polysaccharide in vitro antioxidant activity,the hypoglycemic activity and the repair function for dangered organs as well as the immune activity were studied in the paper,the main result are as follows:(1) On the basis of single factor experiment, for the response surface experiment design and analysis,the best extraction technology were obtained:ultrasonic power200w, temperature90℃, extraction time35min,1:25of material liquid ratio,, under the condition of the available maximum extraction yield is6.94%under the chosen condition.(2)After DEAE-52and SephadexG-100separation and purification of acid crude polysaccharide,seven kinds of polysaccharide PAVF Ⅰ-α、PAVFⅠ-b、PAVF Ⅱ、PAVF Ⅲ、PAVF Ⅳ-α, PAVF Ⅳ-b、PAVF Ⅴ, parts include were obtained.High performance liquid chromatography and uv scanning, show no nucleic acid and protein, infrared spectrum analysis concluded that polysaccharides were for pyranoid sugars and own beta glycosidic bond type of purified four parts polysaccharide PAVFⅡ-α、PAVFⅡ-K PAVF Ⅲ-b、PAVFⅣ.(3) The experimental results indicate that crude polysaccharide has certain antioxidant activity, the scavenging activity of DPPH·,·OH, O2-· are80.77%,57.08%and37.75%respectively at2mg/mL. In addition, the calyx physalis coarse polysaccharide has certain reducing power.(4) On diabetic mice experiment research showed that crude polysaccharides have hypoglycemic function, In the experimental concentration range, High doses crude polysaccharide can be more effective in reducing blood sugar of diabetes mice, reduction was20.96%.(5) In the experimental concentration range, On high dose group of diabetic mice, urea nitrogen, creatinine, alanine aminotransferase, aspertate aminotransferase levels were significantly decreased, Respectively was42.4μmol/L、12.5mmol/L、123u/L、285mmol/L.So Calyx physalis coarse polysaccharide had certain repair diabetes mice liver and kidney function.(6) Crude polysaccharide can enhance the immune organ index, the phagocytosis of macrophage, increase the half hemolysis value and the serum immunoglobulin levels of diabetic mice, so as to improve the immunity of diabetic mice. In the experimental concentration range, the spleen index, thymus index, carbon clearance index, phagocytosis index of the high dose group of diabetes mice was38.51mg/10g、10.19mg/10g、0.036、6.04,and half hemolysis value was68.5, Serum immunoglobulin level was5.19g/L.
