objective To investigate the effect and mechanism of 5-Allyl-7-gen-difluoromethylenechrydsin(ADFMChR) on proliferation and apoptosis in ovarian cancer cell lines COC1 and COC1/DDP cultured in vitroMethods One Chinese Hamster ovarian cell line(CHO-Kl) and two ovarian carcinoma cell lines ,one sensitive (COC1) and the other resistant(COC1/DDP) to cisplatin were used in this study . After being treated by Various concentration of ADFMChR for 24 , 48 , 72hours, Cell proliferation was measured using 3-(4,5-dimethylthiazol-2-yl)-2, 5-dipheny tertrazolium blue (MTT) colorimetric assay . Cell cycle and apoptotic percentage were detected by PI staining flow cytometry . Expression of intracellular Bcl-2, COX2, NF-κB and PPARγ protein was analyzed by indirect immunofluorescent flow cytometric techniqueReSUltS ADFMChR inhibited significantly growth of COC1 and COC1/DDPcells in a dose-dependent manner and a time - dependent manner. After treatment with ADFMChR , the enhanced G1 arrest was accompanied with the augmented apoptotic percentage in COC1, the enhanced S arrest was accompanied with the augmented apoptotic percentage in COC1/DDP, as well apoptotic percentage in two ovarian carcinoma cell lines was significantly higher than that in the normal ovarian cell line (CHO-K1).
