Background: Our previous studies showed that chrysin was taken as leader compound to design and synthesize a series of derivatives: 7-difluorinmethylene -5-substitutied-alkoxyl flavonoid compounds,which all had proliferation inhibitory effects of human gastric cancer cells . Among all the compounds,7-difluorinmethylene -5 -allyl flavone ( DFMeAlChR ) was the strongest .In this experiment , DFMeAlChR was taken as target compound to investigate the proliferation inhibitory effect of human gastric cancer cell line SGC-7901 in vitro and to explore its antitumor mechanism.Method: MTT assay was used to test the proliferation inhibitory effects of humangastric cancer cells (SGC-7901) by DFMeAlChR. Trypan blue staining assay was used to count the number of viability cells,to draw cell growth curve and to inverstigate the growth inhibitory effect of SGC-7901 cells by DFMeAlChR. Plate-colon assay was used to test colon formation inhibitory effect of SGC-7901 by DFMeAlChR .Acridine orange staining was used to observe apoptosis induced function of DFMeAlChR to SGC-7901 cells .PI staining flow cytometry (FCM) was used to analyse the apoptosis rate after treatment with DFMeAlChR for 48h .The protein level and activity of NF-κB,Bcl-2and Bax were examined by Western blot to explore the molecular mechanism of anti-gastric cancer action of DFMeAlChR .Result: The MTT assay showed that the proliferation inhibitory rate was 3.1%, 8.0%, 17.6%, 29.4%, 39.0%respectively after SGC-7901 cells treated with various...
