Objective: The study was to investigate the effects of rosmarinic acid on the PC12 cells apoptosis inducd by glutamic acid and explore the involved underling molecular mechanism.Methods: With the model of PC12 cell injury induced by glutamic acid, the cell shape and nuclear morphology were observed by light microscope and nuclear staining with acridine orange (AO), respectively. The relationships both dose-effect and time-effect of rosmarinic acid on survival rate of the PC12 cells were determined by 3(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT). The effect of rosmarinic acid on apoptotic rate of PC12 cells was determined by flow cytometry analysis dyed with propidium iodide (PI). RT-PCR, indirect immunofluorescence and western blot were used to detect the effect of rosmarinic acid on Bcl-xl, Bax mRNA and protein expression in PC12 cells, respectively.Results: After PC12 cells incubated with deffrent dose of glutamic acid (1, 5, 10, 12.5, 15, 20mmol/L) for 24 hours, the survival rate of cells was reduced in dose- dependent manner.
