Experiments were performed to observe biologic activities of Potentilla Anserine Polysaccharide(PAP). First of all, PAP was extracted and isolated from the root of Potentilla anserine L.. PAP was also purified in terms of its chemical character for the next study. PAP is composed of glucose,arabinose,galactose and rhamnoset. All monosaccharide connected withα-indican bond. Secondly, the basic analyses on the structural features of PAP were studied by ultra-violet absorption spectrum and three chemical reactions. Finally, the effects of PAP on proliferation and function of cultivated immunity cells (lymphocyte, macrophage) from mouse were observed through several immunological tests.The effect of PAP on cells proliferation was evaluated by the method of MTT. In the experiments on signal transduction, sodium nitroprusside (SNP) was selected as a positive control agent. [ Ca2+] concentration was determined by Confocal laser scanning microscope. The variation of NO was measured by the method of Griess. The release of IL-2 was also valued through cells proliferation. In addition,Zymosan was used to stimulated neutrophil cells and created respiratory burst. The mensurations of·OH,O2-·and H2O2 were carried out by three different kits,respectively. The results showed that there were no vitriol radix in the structure of PAP. PAP could accelerate splenic lymphocyte to proliferate more lymphocyte than normal control group and secrete more IL-2. PAP presented an opposite effect on [ Ca2+]concentration in lymphocyte and macrophage. The PAP promoted [ Ca2+]concentration in splenic lymphocyte,but reduced them in celiac macrophage. PAP also offered different effects on the secretion of NO in lymphocyte and macrophage. When treated by PAP,the splenic lymphocytes excreted more and more NO within 48 hour. To the contrary,the macrophage inclined to produce less NO gradually. PAP might adjust the celiac neutrophil respiratory burst induced by Zymosan in resisting O2-·in mouse,showing an obvious capacity of resisting O2-·which had a direct correlation with the PAP dosage. however,it had no obvious effect on·OH.From above results,it was concluded that the PAP developed immunity through cells proliferation and signal molecules and free radicals such as Ca2+,NO,O2-·,However, the relationship between them need to be discussed future.
