The Expression Of MMP-2 And TIMP-2 In Rabbit Carotid Artery Atherosclerosis Plague

Objective:Carotid artery atherosclerosis (CAS) is the common reason of ischemic stroke which can be caused by the debris of the plaque or the mural thrombus or the vesselstenosis itself. The including standard of percutaneous transluminal angioplasty and stenting (PTAS) for CAS stenosis is that≥50% symptom related stenosis or >70% no symptomatic stenosis or <50% symptomatic stenosis with an ulcer plaque. Why shouldn't the≤70% no symptomatic stenosis need PTAS treatment? Studies show that there is not a complete correlation between the size of the atherosclerosis plaque and the degree of the stenosis. More than one half of the plaque ruptures derived from the stenosis which is less than 50%. Most of the vulnerable plaques have a feature of less stenosis and no calcification. The deposition of the vascular smooth muscle in atherosclerosis plaque can be regarded as reparation reaction to the impairment. The failure of the reparation reaction may lead to the plaque rupture. We used airing drying and high cholesterol and lipoid diet to establish rabbit carotid artery injury model, used immunohistochemistry to detect the formation of the VSMC derived foam cell, the expression of MMP-2,TIMP-2,ɑ-SM-actin,apoptosis and proliferation, trying to presume the change of the stability during the progression of the atherosclerosis plaque.Methods: Established rabbit carotid artery injured model by using airing drying and high cholesterol and lipoid diet, 36 rabbits were divided into normal group, injured group (in which the rabbits undwent carotid artery injury and fat diet for 1 month, 2 months and 3 months).ɑ-SM-actin,MMP-2,TIMP-2,apoptosis and Ki67 expression were evaluated by immunohistochemistry. Quantitative dates were expressed by means±SD, comparison between groups were evaluated by t test.
