Background and Aims Corneal neovascularization (CRNV) is one of the severe complications of cornea disease. Hitherto, there are still no efficient drugs to prevent its development with few side effects. Nordihydroguaiaretic acid (NDGA), as a specific inhibitor of lipoxygenase (LOX), is responsible for the inhibition of tumorous and nontumorous angiogenesis. Nordy is a chiral compound based on the chemical structure of NDGA, which is synthesized in our laboratory previously and remaining the antiangiogenesis effect of NDGA without its side effect. It has been confirmed that systemic application of Nordy can suppress the development of diabetic retinopathy through by inhibiting retinal neovascularizaton of retinal. This study is designed to observe the effect on corneal neovascularization, corneal opacity and corneal epithelial wound healing after dropping Nordy in the corneas of Long-Evans rats after alkali injury with three concentrations, and to investigate the inhibitory mechanism of Nordy eye drop with the optimal concentration to provide theoretical foundation for clinical application in the future.Methods1. 0.025%, 0.1%, 0.4% Nordy eye drops that were fulfilled with the criterion of pharmaceutics were prepared.2. The toxicity effects and side effects were observed by praxiology, slit-lamp biomicroscope, light microscope and transmission electron microscope, after 21days at first drop to normal Long-Evans rat's cornea with 0.025%, 0.1%, 0.4% Nordy eye drops.3. Inflammatory corneal neovascularization models induced by alkali cauterization were made in Long-Evans rats which were distributed randomly into 5 groups: negative control group of Nordy solvent, treatment groups of 0.025%, 0.1% or 0.4% Nordy eye drop and positive control group of 0.1% Dexamethasone sodium phosphate eye drop.
