Objective:The purpose of this study is to investigate the expression of intercellular adhesion molecule-1(ICAM-1) in the blood-brain barrier (BBB) and soluble intercellular adhesion molecule-1(sICAM-1); leukocyte infiltration and brain edema and study the effects of hypothermia therapy on them .Then to discuss the mechanism of protection of hypothermia therapy to blood-brain barrier following traumatic brain injury in rats.Methods:Modified Feeney's rat free-falling model was used in the study.60 male SD rats were randomly divided into 4 groups : sham group(n=5),sham-operation group(n=5) , and traumatic brain injury (TBI)group(n=25), hypothermia therapy group(n=25) .Sham operation group : Rats were only made an hole in cranialbone and given no hitting. TBI group: rats were hit on lobi parietalis to induce contusion model. hypothermia therapy group: rats were cooled rapidly to 32.5士0. 5℃for 3 hours after injury. The post 2 groups were parted again into five sub-groups which are redivided into 8h,24h,48h , 72h and 96h four timephases. 2ml peripheral blood was drown-off and centrifuged then took the supernatant after corresponding time .The level of sICAM-1 was measured serially with enzyme linked imminosorbent assay (ELISA). At the same time , the rats were killed and cerebral tissue was taken from perifocus . Immunohistochemical technique was used to examine the expression of ICAM-1, leukocyte infiltration and brain edema were measured by HE staining technique.
