Centrosome of animal cells is a small nonmembranous organelle. centrosome in addition to their role as the microtubule organizing center, plays an important role in regulation of cell life. Centrosome instability is probably associated with tumorigenesis. Now a few centrosome-associated proteins that regulate centrosome functions and maintain centrosome stability were found. However, up to now there has been still little knowledge about the mechanism(s) for this critical biological event.Purpose: BACP gene, a newly unknown functions gene, locates in 20p11.22-p11.1. The previous studies demonstrated that BACP located in the centrosome and interacted with BRCA1 protein. we transfected BACP expression vetor into KYSE 150 cell, and screened one isogenic subline containing over-expressed BACP protein, we examined the difference of biology,anti-apoptosis ability and tumor growth of nude mice between two cell lines. We hope to explore the relation between BACP and tumor.Methods: 1.We transfected BACP expression vetor into KYSE150, an esophageal cancer cell line. We analyzed the BACP protein in transfected cell by Western blotting and fluorescence microscopy, and set up isogenic subline that over-expressed BACP protein. We examined the difference of growth, transwell and adhesion ability between the two cell lines. 2. Two cell lines were treated with UV and cisplatin respectively, then we examined the cleavage of PARP-1 and caspase-9 by western blotting; examined the nuclear morphological changes by immunofluorescence; examined anti-apoptosis ability by flow cytometry and cloning culture. 3.
