Objective : To investigate the relationship between the expression of neutronphil elastase (NE) and monocyte chemotactic protein-1 (MCP-1) in the development of acute lung injury.Methods: To establish a rat model of ALI by intravenous administration of oleic acid (OA) into the jugular vein of male SD rats after one hour of ischemic reperfu- sion of the hind limbs.Forty-eight male SD rats,weighing between 180 and 250 g, were randomly divided into normal saline (NS) group and model group.The model group were randomly divided into group A B C D and E.An equal number of animals in each group were sacrificed after injection of NS or OA at 1hr, 3hr, 6hr, 24hr and 48hr.The level of NE and MCP-1 in blood serum and broncho alveolar lavage fluid (BALF)was measured by ELISA.Results:1. Compared to NS group,After the injection of oleic acid into the oral lip of rat appears cyanosis and their frequency of respiratory turns from 68±14/min to 116±20 /min (P<0.05)2. The histological changes in injury group show that diffuse alveolar damage, inflammatory cells,especially PMN,were found increasingly in capillaries, interstitial tissue and airspaces. Interstitial edema and alveolar haemorrhagia can be foundThe group can be observed local hemorrage and focal infarct form.3.
