The Rat Bone Marrow Stromal Cells Were Induced Into Dopaminergic Neuron By GDNF And GM1 Etc In Vitro

Background and purpose: Parkinson's disease is a frequent dyscinesia disease.It has not specific therapy. In recent investigations,cell transplantation and gene therapy have been the most prospective way, but it is important to be resolved to explore the sources of ideal transplantation cells. Bone marrow stroma not only exist hematopoetic stem cells,but also exist a subset of non-hematopoietic cells referred as marrow stromal cells(MSCs).These cells can transdifferentiate into multiple cell lineages including osteocytes, chondrocytes, adipocytes and so on. The multipotential of MSCs, their easy isolation, culture, proliferation and being able to auto-transplant make them ideal engineering cells in cell and gene therapies for central nervous system diseases.Now people always use different cytokines to control the differentiation of MSCs in vitro,and then use for transplantation when it proliferates to enough cell number. MSCs can differentiate into neuron-like cells by using inductor in vitro,but the inductivity is not high, or the neuron-like cells have not basal function of normal neurons.It is necessary to research a way of high inductivity that MSCs differentiate into normal neuron with function in vitro before clinical application.Our experiment use glial-derived neurotrophic fator(GDNF) and monosialoganglioside(GM1) etc to induce MSCs to differentiate into dopaminergic neuron,our purpose is to approach a way to induce MSCs to differentiate into dopaminergic neuron in vitro,supply a ideal cells source for treating Parkinson's disease and do the ground work for cell and gene therapy.
