Objective To discuss the expression and significance of CDNF,a dopaminergicneurotrophic factor,in animal models with Parkinson’s disease(PD),And to probe theinhibition of CDNF in6-OHDA-induced apoptosis effect.Methods①PD models were fixed with the stereotactic apparatus through themicroinjection of6-OHDA into the left substantia nigra pars compacta(SNc) andmedial forebrain bundle(MFB) of the rats.After four weeks,apomorphine wasinjected intraperitoneally to screen successful models.Through western blotting andimmunohistochemistry detect the expression of CDNF in the substantia nigra of PDmodels.②The CDNF been transfected to MSCs by Lip2000.After the screening byG418,MSCs will be stabilize expression CDNF(also calls CDNF-MSCs cells).Through the MTT analysis and Annexin Ⅴ-FITC cell apoptosis analysis detects thesupernatant to6-OHDA induction of PC12cell apoptosis.Results①The expression of CDNF was low in the substantia nigra of PD models,but it has high expression in control group.②Through the screening by G418,MSCs transfected cells will stably expressCDNF.The supernatant of CDNF-MSCs which have a inhibition effect for it pretreatment of PC12cells,can effectively reduce the rate of apoptosis.Conclusion①The hypoactive of CDNF proclaims that it is playing an importantrole in PD models,which might afford a new therapeutic schedule in clinical.②CDNF which is a secreted protein will striking reduce the6-OHDA-damagedPC12cells.
