AIM: To separate and purify the rabbit VX2 carcinoma cell line in vitro, and try to find if tumor cell invasion is suppressed.METHODS: VX2 tumor fragments were isolated and cultured with explant culture technique and enzymatic digestion in vitro. After 40 passages, the cell morphology, cell cycle, karyotype and tumorigenecity in rabbits and nude mice were investigated. Control group: rabbit VX2 cells were cultivated at 37℃. Experimental group: cells were heated to 43 ℃ for 1 hour. MTT assay was adopted to evaluate cell proliferation after hyperthermia. Invasion test was also put into practice to find if the invasive ability of rabbit VX2 cell line changed. Applying streptavidin biotin complex (SABC) immunohisto -chemical technique, the expression of MMP-2 and VEGF in this cell line was studied.RESULTS: The newly established cell line VX2 was maintained in continuous culture over 50 passages for 8 months. Morphologically, VX2 cells were uniform, round and polygonal epithelial cells. The cells were small but had a high nucleocytoplasmic ratio. The cell cycle analysis showed 74.61% in G1 phase, 7.78% in G2 phase and 17.6% in S phase. The population doubling time was 26.4 h. The chromosomal analysis showed a hypo-triploid with a...
