ObjectWith techniques of 2-DE (Two-dimensional gel electrophoresis), the thesis analyzes the proteome of myasthenia gravis hyperplastic thymus tissue and screens the MG related proteins, which then identified by matrix assisted laser desorption ionization time of flight mass spectrometry (MALDI -TOF-MS).Methods(1) Composed by combining with the second dimension using 12% SDS polyacrylamide gel electrophoresis, the repetitiveness of the 2-DE system was analyzed among the sample proteins of the hyperplastic thymus tissue from 3 patients with myasthenia gravis (MG). 2-DE with the first dimension isoelectrofocusing using 17cm pH3-10 IPG ready strips was employed to compare with the two different modes made up of the sample lysate I and II. Other factors such as method and reset of eletric parameters, protocol for staining and drying the gels were also improved.(2)Using the proper methods described above, satisfactory 2-DE maps of hyperplastic thymus tissue were obtained. Compared with the control (normal serum) and AID (autoimmune disease serum), MG serums were incubated with protein spots on the NCM.
