Aim Chronicity is a prominence character of HCV infection. some extra-hepatic tissue become the resort for HCV replication and storage,it makes against the clearance of HCV. It is significance to explore the role in chronicity of extra-hepatic cells, especially peripheral blood mononuclear cells ( PBMC) with HCV infection in hepatitis C. Now it was thought that injury of liver was caused by immune response, the immune response decided the course and evolution of hepatic C. Human leucocyte antigen (HLA) complex may regulate immune response, and there are thought much of association between HLA and HCV infection. HLA-â…  and â…¡ molecules are ligands of CD8 and CD4 molecules, it enhangce the joint between immunnical cells and reinforce immune response. some HLA genotype might influence the immune response and outcome of infection with HCV . Recent study shewed that HLA-DRB1*1301, 1302 is involved in the clearance of HCV. It is significance to study the effect of HLA-DRB1*1301, 1302 allele on HCV infection and chronicityMaterials and methods Nested polymerase chain reaction (PCR) was applied to detect HCV RNA in the sera and in PBMC, agarose electrophoresis and DNA Marker were applied to ensure the accurate PCR products. And the specific primer of HCV RNA was desighned by software based on sequence in gene bank. Genomic DNA was extracted from white blood cells of patients using traditional...
