Study On The Isolation And Bioactivity Of Active Ingredients From Chinese Traditional Herbs Reheum Officinal Baill And Ribes. Henry French Et Al.

To seek for bioactive products from herbs in Guizhou Province, nine plants of eight families were primarily studied for the bioactivity of their crude extracts. The plants include Pinellia ternata (Thunb) Brief, Medicinal Evodia Fruit, Hypericum japonicum Thunb, Herba Senecionis Scandentis., Astragalus membranaceus (Fisch.) Bge., Herbal Houttuynia, Pueraria Lobatae Root., Reheum officinal Baill, Ribes. Henry French. During the course of research, two medical plants Reheum officinal Baill and Ribes Henry French were found highly active against plant fungus (Gibberella zeae, Fusarium oxysporum, Phytophthora infestans) and were studied systematically. Fifteen ingredients were isolated from the two plants and identified by spectroscopic and chemical methods. Among them, eleven ingredients were isolated from Reheum officinal Bail and ten structures of them were established by IR, MS, ¹H NMR and ¹³C NMR.. Four compounds were isolated from Ribes Henry French and two of them were established by IR, MS, ¹H NMR and ¹³C NMR.The natural hydroxyanthraquinones, physion and emodin which are effective ingredients of Chinese traditional medicine exhibited antitumor activities against PC3 cells and A431 cells.It is the first time to report here the inhibitory activity against EGF (epidermal growth factor)phosphorylation of five hydroxyanthraquinones from Rheum. Compound 4 (emodin) hasinhibitory activity against ERK (extracellular signal-regulated kinase) phosphorylation in PC3(prostate cancer cell lines) cells induced by EGF which was concluded to be a potential ERKphosphorylation inhibitor.Crude extracts from Reheum officinal Baill showed good activities against fungus. However, no activity was observed for the purified compounds from crude extracts. It was possible that some compounds in the crude extracts act corporately. Therefore, a test on mixtures of different ingredients was designed to prove the supposition reasonable.To the best of our knowledge, this is the first research on the extraction and isolation of Ribes. Henry French, root. And it was found that the crude extracts possess moderate to good antifungal activity but weak activity against TMV (tobacco mosaic virus ) .
