Objective: Cytokinesis-Block micronucleus method was used to study the chromosome injury on the SHG44 cells treated by 60Coγ-rays.Impacts of cell survival,apoptosis and cell cycle progress induced by Wortmannin(WORT), and by WORT combined with 60Coγ-rays in the SHG44 cells were studied. Using gene transfection technique, the AKT plasmid was transfected into the SHG44 cell in which we aimed to study the influence of AKT on the radiation effect of proliferation and apoptosis, in all to prove the relation of AKT with radioresistance of the SHG44 cells.Methods : Using Cytokinesis-Block micronucleus method , the micronucleusfrequencies(MNF) and micronucleus cell frequencies(MNCF) of SHG44 cells withexposure to 60Co γ-rays were observed. Cell survival was measured by MTT( Methylthiazolyl tetrazolium) assay and by colony formation. Cell cycle progress was investigatedby Flow cytometry. Morphology features of apoptosis were determined by laser confocalmicroscope in SHG-44 cells stained with AO/EB. Electroporation was used to transfectAkt into SHG44 cells, exogenous expression of AKT mRNA was detected by RT-PCR.Results: (1)Both MNF and MNCF increased with radiation dose, a small shoulder wasobserved in a small dose range from the curve. At 3 Gy, both MNF and MNCF promptlyincreased.(2)Concentration-dependent decrease of cell survival rate was induced byWORT form 1.00×10-8 to 5.00×10-7mol/L, no evident change was observed from5.00×10-7 to 5.00×10-6mol/L,and another decrease was observed from 1.50×10-5 to3.00×10-5mol/L. Dose-dependent decrease of cell survival rate was induced by60Coγ-rays,interaction between 60Coγ-rays and WORT was observed. Increase of thenumbers of cells in Go/Gi and S phase but decrease of the numbers in G2/M phase wereinduced after exposed to 10Gy, the increase of numbers of cells in G2/M and S phase butthe decrease of the numbers of cells in G0/G1 phase were induced by 0.5 ×10-5mol/LWORT.
