Radiotherapy is one of the major treatment modalities for cancers. But the radioresistance of some cancer cells has a significant effect on the treatment results. It has been founded that many genes have a relationship to the formation of radioresistance. But the mechanism is not quite clear. Before this study, the gene expression in the radioresistant variant human esophageal cancer cell line TE13R120 and its parent cell line TE13th had been examined and expression difference was found among 57genes, in which the expression difference of HDAC3, NRAGE and NF-kB was increased with the prolonged time between the two cell lines. The study was designed to verify the reliability of cDNA microarray on detecting the gene expression differences between the radioresistant variant human esophageal cancer cell line TE13R120 with its parent cell line TE13, to detect the mRNA and protein expression differences of gene HDAC3, NF-kB, NRAGE in the two cell lines, and to investigate their association with the formation of radioresistance of TE13R120 cell line. In addition, a study was done about the effect of Trichostatin A (TSA) on the HDAC3 expression, cell proliferation, radiosensitivity, cell cycle distribution and apoptosis of the two esophageal cancer lines with different radiosensitivities.Part OneObjective:The study was designed to verify the reliability of cDNA microarray on detecting the gene expression differences between the radioresistant variant...
