Aim: To evaluate the effects of osthole on fatty liver, and investigate the possible mechanism.Methods: Quail model with hyperlipidemic fatty liver and rat model with alcoholic fatty liver were set up by feeding high fat diet and alcohol, respectively. These experimental animals then were treated with osthole 5—20mg/kg for 6 weeks. The blood-lipid, hepatic-lipid, coefficient of hepatic weight, SOD, GSH-PX, MDA and liver histological changes were observed. Expressions of CYP7AmRNA, DGATmRNA and PPARamRNA in hepatic tissue of alcoholic fatty liver rats were examined by RT-PCR method.Results: After treated with osthole 5-20mg/kg for 6 weeks, the levels of serum TC, TG, LDL-C, coefficient of hepatic weight, and the hepatic tissue contents of TC and TG were significantly decreased (P<0.05 or P<0.01) , and the activity of SOD in liver was improved (P<0.05 or P<0.01) . In alcohol-induced fatty liver rats, level of MDA in liver was decreased (P<0.01) . In high fat-induced fatty liver quails, GSH-PX in liver was significantly improved (P<0.05 or P<0.01) . The histological evaluation of liver specimens demonstrated osthole dramatically decreased lipid accumulation (P<0.05 or P<0.01) . Osthole could significantly increase the expressions of CYP7AmRNA and PPARamRNA (P<0.01) , decrease the expression of DGATmRNA in hepatic tissue of alcoholic fatty liver rats (P<0.01) .
