Study Of B7 Costimulatory Molecule Regulating Liver Egg Granuloma Formation And Its Molecule Mechanism In Mice Infected With Schistosoma Japonicum

In this paper, we probed the associativity between Th1/Th2 immunodeviation and egg granuloma in mice infected with Schistosoma japonicum, based on which we observed in vivo and in vitro the regulating effects of B7-1 active mAb on Th1/Th2 immunodeviation and on egg granuloma formation, and probed the molecule mechanism of its immunopathology. Our results showed that interference of B7:CD28 costimulotary pathway with B7-1 active mAb could significantly up-regulate the expression of Th1 cytokine IFN-γ(P<0.05),while the expression of Th2 cytokine IL-4 was not obviously affected. Analysis of IgG2a/IgG1 and Th2 differentiation index, which can reflect Th1/Th2 immunodeviation in a way, indicated that B7-1 active mAb could mediate Th1/Th2 immune response to the Th1 deviation. A marked decrease in the size of egg granuloma also occurred in mice treated with mAb (P<0.05). Immunohistochemistry showed enhanced expression of B7-1 and reduced expression of B7-2 on the hepatic granuloma of mAb-treated mice. Expression of NO in supernatant of splenic lymphocytes was in positive correlation with that of IFN-γ,and in negative correlation with that of IL-4,indicating NO might be another important factor in regulating the pathology of egg granuloma. Our research showed that interference of B7:CD28 costimulotary pathway with B7-1 active mAb could regulate egg granuloma formation by mediating Th1/Th2 immune response to the Th1 deviation, thus provided theoretical base for a novel way to control egg-granuloma formation.
