Study On The Mechanism Of Octylphenol-Induced Toxicity In Cultured Rat Testes Sertoli Cells | Posted on:2007-03-30 | Degree:Master | Type:Thesis | Country:China | Candidate:J Qian | Full Text:PDF | GTID:2144360185479159 | Subject:Occupational and Environmental Health | Abstract/Summary: | PDF Full Text Request | Part I Study on the Apoptosis Induced by OP in Cultured Rat Testes Sertoli CellsIn order to explore the apoptosis induced by OP in raits testis Sertoli cells and its mechanisms, we selected adolescent male rats (25-30d) to set up Sertoli cell primary culture model in vitro, and directly treated Sertoli cells with 30-60 μM OP for 24 h or 60 μM OP for 6-24 h. We identified the OP-induced apoptosis by transmission electron microscope, flow cytometric analysis and Hoechst staining. RT-PCR was used to analyze the levels of Bcl-2 and Bax mRNA. Bcl-2, Bax and caspase-3 protein expressions were determined by western blot analysis. The main results read as follows:1. Examined by TEM, the control cells had normal Sertoli cell morphology with abundant subcellular organelles in the cytoplasm. The nuclei had uniformly dispersed chromatin, clear nucleolus and satellite nuclear bodies. After treatment of OP, Sertoli cells were mainly characterized by irregular shapes, condensation of nuclei,...
