Bone defects are formidable clinical challenges of orthopaedics, which often secondary to tramua, tumors, osseous infection, and fracture nonunion. Recently, great development in tissue engineering has provided a novel cue for the management of critical large bone defects. As seed cells of bone tissue engineering, bone marrow-derived mesenchymal stem cells are multipotential cells capabl of differentitating into osteoblasts, chondrocytes, adipocytes, and other connective tessues cells. In this experment, we choose allogenic bone marrow mesenchymal stem cells as seed cells and fibrin as scaffolding materials and observe the compound grafts' curative effect of repairing cancellous bone defects in rabbits.Objective To discuss the isolation, culture of bone marrow MSCs in vitro and the curative effect of repairing cancellous bone defects in rabbits with the comound of allogenic MSCs and fibrin. To investigate immunological rejection evoked by allogenic MSCs.Methods MSCs were isolated and culture-expanted from rabbit bone marrow. Cancellous bone defects were created on bilateral femur condular in 15 Japanese big ear rabbits, the left bone defects were experment lateral, which were treated with compound of allogenic MSCs and fibrin, the right were control lateral, which were implanted with fibrin only. The bone defects were examined radiologically and histoloyically at the 2nd, 5th, 8th week after operation.Results Radiologically, the bone defects treated with the compound of MSCs and fibrin implantation had a little bone formation within 2 weeks.
