Objective To study the protective effect of XNN on cerebral ischemia injury and itsmechanism.Methods (1) The experiments of quick decapitation and anoxia under normal pressure model in mice was used to study the effect of XNN on acute cerebral ischemia and anoxia. (2) The global cerebral ischemia /reperfusion model was established to survey the influences of XNN on the recovery time of righting reflex and electrical activity of brain , the water content and cerebral index , the activity of SOD , LDH, and NOS and the content of MDA, LD in brain tissue and ET in hippocampus . (3) The focal cerebral ischemia reperfusion modal was made to observe the effect of XNN on neurological function score, infarction volume of brain, thrombogenesis in vitro, platelet aggregation, tumor necrosis factor- α , pathomorphology of brain and the ultrastructure of neuron in rats. (4) Lysing effect of XNN on blood clot was observed in vitro. (5) The effect of XNN on cerebral blood flow , cerebral vascular resistance , blood pressure, heart rate and electrocardiographic were detected in dog.Results (1) XNN (1 5,30g/kg) could prolond the gasping time of mice after quick decapitation and XNN (30g/kg) could prolond the survive time of anoxic mice. (2) The globe cerebral ischemia reperfusion experiment induced by four vessels occlusion(4VO) for 10min and reperfusion for 24h was performed to evaluate effect of XNN on acute neuronal injury in rats. During reperfusion after ischemia, XNN (6, 12g/kg) significantly promoted the recovery of electroencephalogram and righting reflex. XNN(6, 12, 24g/kg) could reduce the brain edema and brain index obviously. SOD,LDH activities increased and MDA,LD content decreased in brain homogenate in rats treated by XNN (6, 12g/kg) . XNN might inhibited the NOS activity in brain homogenate and ET content in hippocampal homogenate in rats.
