Objective : To determinate the content of the active components in shenqin compound prepration, study the apoptosis of HSCs induced by shenqin compound preparation. Method: To determinate the content of the baicalin, baicalein, Tashinone I , TashinoneIIA by RP-HPLC. In vitro study,we investigated the effect of different concentration shenqin compound preparation (10μg/ml, 30 μ g/ml, 50 μ g/ml, 100 μ g/ml, 200 μ g/ml) on rat hepatic stellate cells (HSC)and hepatic cells(HC) by MTT. In vivo study, two rat liver injury models induced by CCL4 and bile duct ligation, investigated the effect of rat serum AST , ALT, HYP, HA, LN by low dose (20 mg/kg) , middle dose (50 mg/kg ), large dose (100 mg/kg) of shenqin compound preparation, and observed the change of liver tissue pathology .Result: The linear range of baicalin, baicalein, Tashinone, TashinonellA were well, r=0.9992, r=0.9992 , r=0.9994, r=0.9993, While the average recovery rate were exceed 97.24%.In vitro study ,HSC were exposed to 10 -200 μ g/ml shenqin compound preparation caused a dose -dependent reduction on cell viability and no injury to HCs. In vivo study , shenqin compound prepration (large, middle dose) significantly reduce the levels of serum AST, ALT, HYP, HA, LN ,significantly different from model group, and the observation of pathology indicate the shenqin compound prepration can reduce the injury of rat liver tissue. Conclution: The method of the RP-HPLC is rapid, accurate and reliable,the vitro study indicate shenqin Compound prepration could promote the apoptosis of HSCs and no injury to HCs, the vivo study indicate shenqin compound preparation could improve liver fibrosis and injury by ccl4 and bile duct ligation in rats.
