The purpose of this research is to study Pharmacokinetics and Pharmacodynamics of Compound HuangBo Granule. Berberine of Compound HuangBo Granule was determined by RP-HPLC assay and it can be used for quality assessment; Pharmacokinetics of Compound HuangBo Granule in mice and the Pharmacokinetic parameters were investigated; different in vitro and in vivo experiments were designed to test its various pharmacological effects. These experiments were consisted of three parts as follows:1. Reverse Phase High performance Liquid Chromatography(RP-HPLC) Determination of Berberine in the Compound HuangBo GranuleThe RP-HPLC assay was invested to determine the content of hydrochloric berberine in Compound HuangBo Granule. The chromatogram condition was optimized as follows: Hanbon-C18 column which is 4.6mm × 250mm and 5 u m as a stationary phase, acetonitrile-water(50:50, KH2PO4 3.4g and SDS 1.7g in 1000mL solution) as a mobile phase, eluent rate of 1 mL/min, detection at 346nm, temperature of column is 30℃. Berberine was quantified with external standard method. The experimental result showed Berberine was completed separated form its impurities and a good linear relationship between the peak area and mass concentration of the berberine within the range of 3.0 - 15.0 μ g/ml(r=0.999749). The relative standard deviation(RSD) was 2.04%(n=6). The average recovery was 100.25%. The content of hydrochloric berberine is 0.4385mg/g Compound HuangBo Granule.2. Study on Pharmacokinetics of Compound HuangBo Granule in MicePharmacokinetics of orally administered at a dose of 25 g/kg Compound HuangBo Granule(10mg/kg berberine) were investigated in mice. Serum concentration of the effective component-hydrochloride berberine were measured using HPLC and concentration-time data analyzed with Pharmaceutical Kinetics Software.
