Background and Objective:Dental caries is one of the common infectious diseases of human, and many a study has proved that S.mutans is the primary causative organism of dental caries. Attachment to, and accumulation on tooth surfaces is essential for cariogenicity of S.mutans. Immune defence against dental caries focus on preventing S.mutans from adhering to tooth surface by blocking adhensins and inactivating virulence factors on S.mutans' surface that play important roles in S.mutans' adherence by active or passive immune means. In the nineties of last century, the discovery of DNA vaccine has been praised as the third revolution of vaccinology, much light is recently thrown on caries DNA vaccine. In previous study we have successfully constructed the dual-promoter expression vector harboring SBR gene and proved its immunogenicity. To continue this study we were going to construct prokaryotic expression vector pTriEx-4-GBR containing the Glucan-binding region (GBR) gene of S.mutans GTF-I, then purify the recombinant GBR protein and prepare the multiclone antibody elicited by the recombinant GBR protein; which lay the foundation for further studies including construction and analysis of bivalent dual-promoter caries DNA vaccine.
