Objectives: To investigate the effect of tetromethylpyrazine and aminoguanidine on expression of transforming growth factorβ1mRNA(TGF-β1mRNA), insulin signaling molecules insulin receptor mRNA (InsRmRNA) and protein kinase C β 1 (PKC β 1mRNA) of type 2 diabetic rats ,to explore the therapeutic mechanism of the insulin resistance.Methods : The Wistar rats of 90 in 24 hour were divided into 2 groups:nomal control group (group C)and diabetic group. Diabetic group were induced by streptozotocin, When rats were 2 months old, the intraperitoneal glucose tolerance test (IPGTT) were performed, 2 hours blood sugars >7.8mmol/L were seleted in this research. They were divided into 5 groups: untreated diabetic group (group DM), aminoguanidine treated group (group AG), tertromethylpyrazine treated group (group TMP), tertromethylpyrazine and aminoguanidine treated group (group TMP+AG), metformin hydrochloride treated group (group MH),vitamin C treated group (group Vc), each group of 10. The DM group drinks the running water, each treated group drinks contains the corresponding medicine in the running water, 50mg/Kg /d. By the tail cut method every month for 6 months, the fasting plasma glucose, glucose tolerance test , plasma insulin, plasma glycosylated serum protein (GSP), nitric oxide (NO), reactive oxygen species (ROS), superoxide dismutase (SOD) was detected every month. At last, analysis of the difference of plasma biochemistry and the expression of mRNAs of TGF-β1,InsR and PKC among the seven groups was performed.
