Objective:To isolate,purify and propage mouse bone marrow-derived dendritic cell(DC),investigate the changes of morphological structure and bio-immunological characteristics of DC during its developmet,differentiation and maturation.To construct DC vaccine pulsed with RM-1 prostate cancer cell lysate,detect the specific anti-tumor activity of DC vaccine in vitro and vivo.Methods:The C57BL/6 mouse bone marrow precursor cells were isolate and cultured with cytokines of GM-CSF and IL-4.The morphological character of DC in different stages were studied by using light-microscopy,and the immunobiologic of features were studied by mixed leucocyte reaction(MLR) and horseradish peroxidase (HRP) phagocytosis essay.DCs were pulsed with RM-1 prostate cancer cell lysate, and IL-12 was assay in the supernatant of sensitized DC culture fluid by enzyme linked immunosorbent assay(ELISA).The effect of inducing specific kill activity of DC vaccine was detected. The model of mouse prostate cancer was establish.The therapeutic effect of sensitized DC on mice with prosteta cancer was assessed.Rusults:1.DC were harvested from mouse bone marrow progenitors followed culturing the cells with GM-CSF or GM-CSF plus IL-4 for 7days. After 4 days,immature DC possessed short processed,and many of vacuoles in the cytoplasm.They failed to stimulate allogeneic T cell proliferation,and could phagocytose vigorously HRP granules. After 7 days,mature DC possessed long processed,and few of vacuoles in the cytoplasm.They had activity of stimulating allogeneic T cell proliferation,and could phagocytose slenderly HRP granules.
