Studies On Chemical Constituents And Fingerprints Of Nauclea Officinalis

Nauclea officinalis is an important medicion-used species of genus Nauclea Rubiaceae,which grows in the southern part of China. The stem and bark of Nauclea officinalis are all valuable for medicine. It has been used as an anti-inflammatory and antibacterial agent in Chinese folk medicine. It has been also a kind of often used folk medicine abroad. The genus Nauclea is a rich souce of alkaloids.To date, more than 40 monoterpenoid indole alkaloids have been isolated from this genus.It was reported that some of these alkaloids have various bioactivity, such as antirheumatism activity, antimicrobial activity, antiparasit activity and antimalarial activity etc. There were wide clinical application of troche and injection of nauclea officinalis at hainan province,guangdong province and guangxi provmce.They both have good effect at the treatment of respiratory infection, acute-amygdalae inflammation, bronchitis, urinary infection etd. 90% of them were effective and there were no obvious side effect.Thinking about its good clinical value and various bioactivity, we took a systemic reseach on the chemical components ,assaying and fingerprint of Nauclea officinalis .In the research of the chemical constituents,we used silica gel colomn chromatography, gel colomn chromatography and semi-preparative HPLC, 18 constituents have been isolated. On the basis of their chemical and physical properties, spectral data of ¹H-NMR, ¹³C-NMR, DEPT, ¹H-¹HCOSY, HMBC, HMQC, IR, ESI-MS, UV, TLC, 7 compounds have been indentified,including: (1) Vincosamide; (2) Compound5; (3) Compound1; (4)2,3-dihydroxybenzoic acid; (5) 3,4-dihydroxybenzoic ester; (6) Pumiloside; (7) Daucosterol. Vincosamide is the first time extracted from this kind of plant,and the purity is very high (>99.0%) .Compound5 and Compound1 are both absolutely new componds.Vincosamide is a kind of new alkaloid extracted from N.officinalis for the first time.Thinking of the bioactivity of this kind of alkaloid, it is valuable to develop a assaying method .In assaying on chemical constituents,we developed a rapid, accurate and sensitive...
