Purpose:This experiment using real-time quantitative PCR and Western blotting protein,pre hypertension model detection of salt sensitive rats established in kidney tissue in transforming growth factor beta 1 expression of p38 MAPK mediated signaling conduction pathway related genes and proteins,mainly p38 mitogen activated protein kinase and c AMP response element binding protein,and alpha smooth muscle actin,fibronectin gene expression,to investigate the effect of acupuncture principle of renal fibrosis delay of pre hypertension rats,the occurrence and development of a deeper understanding of hypertension.Material and method:In this experiment,the experimental animals were divided into four groups according to the purpose of the experiment,which were normal control group,model group,acupuncture group and non acupoint group.All the experimental animals were 40 healthy male rats,of which there were only Dahl salt sensitive rats,which were fed with high salt diet.They were randomly divided into three groups according to the random number table method,each group was divided into 10 groups.Another 10 Dahl salt resistant rats,given the high salt diet after the blood pressure stable unchanged.The rats were housed separately in 7 days of adaptive feeding,high salt diet and blood pressure changes every day without exception,the blood pressure in salt sensitive rats increased 10-20 mm Hg after cessation of high salt diet instead of ordinary feed,namely the model for success and start the treatment.The rats in the model group only daily binding 20 min static;acupuncture group acupuncture bilateral Zusanli(ST36)Quchi(ST11)acupuncture on 3-5mm,20 min,using density wave,the frequency with the needle handle slight tremor,1 times a day,6 times a week for five weeks;the non acupoint acupuncture group rats with double side iliac crest 15 mm,posterior midline 20 mm beside the fixed control points,using the same density wave,20 min frequency with acusector,needle handle a slight trembling for the degree,the date of treatment and acupuncture group.Blood pressure was measured once a week during treatment.Stop acupuncture.When the sample was drawn,the rats were injected with 10% chloral hydrate(100g rats after administration of 0.35 ml each),after weighing in accordance with the administration of 0.5ml/100 g,the rats were anesthetized,abdominal incision,full exposure of the kidney after resection of the renal capsule,to cut along the coronal section,the maximum longitudinal,to obtain a part of kidney homogenate extraction of total.RNA of renal tissue,expression was detected by real time PCR in the kidney of TGF-beta 1,p38 MAPK,CREB,FN and alpha-SMAm RNA;the expression of phosphorylated protein by Western blotting detection of renal tissue p38 MAPK,Results: 1.The effect of electric acupuncture on the TGF-?1 content of the rats renal tissue.Compared with the normal group,the expression of TGF-1 gene was increased in the model group(P<0.05).After five weeks of treatment,the acupuncture group and non acupoint group rats compared with model group,TGF-beta 1 gene expression decreased(P<0.05),compared with the non acupoint group,acupuncture group decreased significantly,with statistical significance(P<0.05).2.The effect of electric acupuncture on the p38 MAPK signal transduction pathway of the rats renal tissue.Compared with the normal group,the expression of p38 MAPK and CREB in the renal tissue of model group was higher than that of normal group(P<0.05).After five weeks of treatment,the acupuncture group and non acupoint group rats compared with model group,p38 MAPK,CREB gene expression and phosphorylation protein expression were decreased(P<0.05),compared with the non acupoint group,acupuncture group decreased significantly,with statistical significance(P<0.05).3.The effect of electric acupuncture on ?-SMA?FN content of the rats renal tissue.Compared with the normal group,the expression of-SMA and FN in renal tissue of model group increased(P<0.05).After five weeks of treatment,the acupuncture group and non acupoint group rats compared with model group,the expression of alpha-SMA and FN gene were decreased(P<0.05),compared with the non acupoint group,acupuncture group decreased significantly,with statistical significance(P<0.05).Conclusion: 1.EA bilateral "Zusanli" Quchi delayed pre hypertension rat renal extracellular matrix deposition.2.The mechanism of action may be related to the inhibition of the expression of related genes and phosphorylated proteins in TGF-1 mediated p38 MAPK signaling pathway.3.TGF-1,p38 MAPK,CREB,FN and alpha-SMA are the key points.
