Objective To study the influence of intrauterine hypoxia (IH) to offspring rat Ethology change and nerve cells in brain hippocampus degeneration and apoptosis. Methods S-D rat intrauterine hypoxia models was established, then the offspring were exposed to high-fat diet (HF) or (and) second hypoxia (SH); T maze was used to test the space learning and memory abilities in the rats (swimming right rate and in water time as indices); The numbers of the nerve cells in the rat brain hippocampus were calculated with CMIAS system; The numbers of apoptotic figures (TUNEL technique) were investigated; Microstructure of the nerve cell and microvascular in brain were observed by electron microscope. Experimental data was analyzed by triple pathogens two-levels analysis of variance. Results Main effects of IH and HF could both decrease offspring rats'swimming right rate (main effects: 12.80, 8.63,all P<0.05), meanwhile, there were interactions between IH and HF,IH and SH(P<0.05);And the main effects of IH,HF and SH all could extend the swimming time of offspring rat (main effects: 0.92,0.42,0.89;all P<0.05), interaction was only present between IH and SH; Main effects of IH,HF and SH all increased the apoptosis cells in offspring rat pallium, (main effects: 3.40,3.35,2.10;all P<0.05),and interactions existed between IH and HF,IH and SH; Main effects of IH and SH reduced nerve cells in offspring rat hippocampus (main effects: 0.83,1.04,all P<0.05), and interaction existed between IH and HF,IH and SH(both P<0.05);At the same time , interaction among IH,HF and SH was found(P<0.05). Under the electron microscope, compared with blank group, more apoptosis nerve cells, deposited lipofuscins, swelling chondriosomes and damaged microvasculars were observed in IH group offspring rat pallium. And most serious figures were found in triple pathogens crossing group. Conclusions Intrauterine hypoxia may bring harmful effects to nerve cell of offspring rats , increase the risk of nerve cell degeneration or(and) apoptosis and weaken rat space learning-memory capability by main effects. Moreover , Intrauterine hypoxia may aggravate these changes by interactions when being exposed to high-fat diet and (or) second hypoxia. So Intrauterine hypoxia might increase the risk of offspring nerve cells to suffer diseases .
