Objective:It is well known that both nuclear factor-kappa B transcription system and renin - angiotensin system have intimate relationship with myocardial ischemia-reperfusion injury.Here we observed the changes of NF-κBp65 and AngII during various periods of time and investigated the interaction between NF-κBp65 and AngII. Furthermore,this study was to initially investigate the effects of pretreatment with puerarin on NF-κBp65 and AngII.Methods:Rat models of myocardial ischemia-reperfusion injury were successfully established.1.Rats were randomly divided into 5 groups:sham-operation group,30 min of ischemia followed by reperfusion for various periods of time(15,30,60,120 min). The localization of NF-κBp65 was detected by immunohistochemistry and the quantitation of activated NF-κBp65 was detected by ELISA;the content of AngII in blood plasma and cardiac were determined by radioimmunoassay.2.We pretreated with pyrrolidine dithiocarbamate known as a specific inhibitor of NF-κB ,imidapril known as a specific inhibitor of angiotensin-converting enzyme,and both of them to observe changes of the same indexs.3.We pretreated with puerarin of different does to observe changes of the same indexs.
