Objective To investigate the nephroprotective effect of rosiglitazone on 5/6 nephrectomized rats and explore its mechanism.Methods Chronic renal failure was induced by 5/6 nephrectomy, the nephrectomized rats were randomly allocated to Rosiglitazone treatment group 1(5mg· kg-1·d-1 ) (XL group), Rosiglitazone treatment group2 (15mg ·kg-1·d-1) (DL group), and control group (NX group) .The rats with sham operation served as control (SHAM group) . Then 5mg/kg Rosiglitazone, 15mg/kg Rosiglitazone were respectively delivered daily by gavage for 8 weeks, 24 hours urinary protein, BUN, serum creatinine(SCr), serum total cholesterol, triglycerides, blood pressure(BP), Angiotensin II(A2),erdothelin(ET) were measured at the end of the stuffy. Renal pathological changes were evaluated and immunohistochemistry was used to examine the expression of macrophage chemoattractant protein 1 (MCP-1), matrix metalloproteinase-2(MMP-2) and matrix metalloproteinase-9(MMP-9). Renal mesangial collagen fibers were stained with Picric acid-Sirius red. Renal expression of TGF- β1 mRNA and FN mRNA was detected by RT-PCR.Results(1) Compared with the untreated 5/6 nephrectomized group (NX group), blood pressure of Rosiglitazone treatment groups rats were substantially decreased .(p<0.01).(2) Compared with the NX group, 24 hours urinary protein, BUN, serum creatinine(SCr), serum total cholesterol of Rosiglitazone treatment groups rats were substantially decreased, (p<0.05).(3) Compared with the NX group, Angiotensin II(A2), erdothelin(ET)of Rosiglitazone treatment groups rats were substantially decreased, (p<0.05).(4) Pathohistological study revealed that, compared with the NX group, there...
