Objective: To study whether or not lornoxicam reduced pain in the rat incision model and to discuss the possible mechanism,mainly discuss the mechanism of spinal cord center.Method: 18 awake rats were randomly divided into 6, 6, 6 three groups, the first two of which were intraperitoneal injected bidistilled water and lornoxicam 3mg/kg respectively, the third of which was intraperitoneal injected bidistilled water 20 min prior to the incision in the plantar of the right hind paw. 32 awake rats were randomly divided into 4 groups that also received incision in the same region. To test the dose-dependent manner, 4 groups received lornoxicam (0.33, 1, 3, 9mg/kg) 20 min prior to the incisions.In the behavioral study, stepping behavior was evaluated 0-60 min after hind paw incision. Three hours later, rats were extracted blood from heart then perfused, L4-L5 sections of spinal cords were dissected, sliced and processed by immunostained with a rabbit polyclonal antiserum directed against c-fos or a goat polyclonal antiserum directed against COX-2. The changes of FLI and COX-2 positive neurons in the dorsal horn were studied. The plasma PGE2 were mensurated by radioimmunology method.
