Objective To investigate the difference of the expression of theproto-oncogene eIF4E between in larynx squamous carcinoma constitution and in benign lesion, to find out the correlation of eIF4E and larynx squamous carcinoma, for the application of clinic, to use eIF4E as a marker of diagnosis, treatment and prognosis. Method Forty Paraffin-embedded larynx cancer specimens were selected to be used to investigate. The average age of the patients who underwent surgery for squamous cell cancers of the larynx is 58 years (range from 39-78 years). Of whom, there are 38 males and 2 females, and there are 2 patients with metastasis of lymph nodes. Twenty polyps of vocal cord specimens were selected to be control. Immunohistochemical (SP) analysis of all the specimens with polyclonal antibody to eIF4E was performed. Gradation of staining depended on intensity of staining and percentage of cells staining. Scores were ranked as staining, weak staining, intermediate staining, and strong staining. Values of 0, 100, 200, 300 were assigned to the intensity of staining. Tumor eIF4E scores were determined by multiplying the intensity of staining value by the percentage of cells staining positive. Result The expression of eIF4E is found in both tumor group and the polyps group. The expression of eIF4E in tumor group and in polyp group has significant difference (t -test, P<0.05). The expression of eIF4E doesn't correlate with the patients' age, and didn't correlate with the pathology grade and clinic scale. There is no detectiveexpression of eIF4E in the metastasis lymph nodes. Conclusion Expression of eIF4E has significant difference between larynx squamous carcinoma and in polyps,and it didn't correlate with the age and the tumor's pathology grade and clinic scale.In a word, eEF4E is only related to that the lesion is malignant or benign.Postgraduate Ge Ruifeng (Otolaryngology)Directed by Zhao Shuyou...
