[Objective] The aim of this study was to investigate changes in the gene and protein expression of BMP-2, MMP-13, collagen type II in human nucleus pulposus of the degenerated lumbar intervertebral discs, to analyze the related significance of them in the process of intervertebral disc degeneration, to supply the theoretical support for treating the degenerative disc disease.[Methods] The experimental group consisted of 30 cases of the fresh pulpy nucleus tissue which were obtained from patients with lumbar disc herniation by surgical intervention. 8 cases of the fresh pulpy nucleus tissue harvested by operation for fracture of lumbar vertebrae acted as control group. The mRNA and protein expression of BMP-2, MMP-13, collagen type II in 38 specimens were detected with in situ hybridization and immunohistochemical technique. By quantitatively analyzing all the sections with graph analysis meter, the related significance of BMP-2, MMP-13, collagen type II in two groups of specimens was explored.[ Results ] (1)The results of in situ hybridization and immunohistochemical staining showed that there were more strong expression of BMP-2 and MMP-13 in experimental group than that of in control group. On the contrary ,there were more strong expression of collagen type II in control group than that of in experimental group(P<0.05). (2)There was certain negative correlation between the mRNA and protein expression of BMP-2 and that of collagen type II in experimental group( r=— 0.602, -0.614; P<0.01). It was the same to MMP-13 and collagen type II ( r=— 0.615, -0.774; P<0.01).[Conclusions] (1) BMP-2 and MMP-13 are two important regulatory factors in intervertebral disc degeneration.
